Tuesday and Wednesday. Mail dropped early in the week moves through the normal weekday flow, and letters mailed Tuesday or Wednesday typically arrive before the weekend. Friday afternoon is the trap: anything that misses the last pickup sits until Monday, so a Friday letter can take longer than one mailed the following Monday morning.

Two things stack against it. First, the last pickup: every collection box and post office has a final collection time, and anything dropped after it waits. Second, letters don't move on Sundays. A letter mailed Friday at 5pm sits Friday night, all of Saturday, and all of Sunday before the Monday sort. A letter mailed Monday at 9am starts moving immediately and can easily arrive first.
This is the single most useful timing trick in the whole postal system, and almost nobody thinks about it: if you miss the Friday pickup, you're often better off holding the letter until Monday morning than dropping it in the box Friday night. The exception is a post office with a late lobby slot, which buys you a few more hours.
For business mail, yes. A letter that arrives Monday morning joins the weekend pile and the Monday meeting rush; the same letter arriving Wednesday lands on a calmer desk and gets read sooner. If you're mailing a proposal, an invoice, or anything you want actually read, midweek arrival is quietly better.
For households it matters less, though there's a small version of the same effect: mail that arrives Saturday sits until someone deals with it, while a Wednesday letter gets opened the same evening. None of this changes delivery speed, but it changes how fast the letter gets attention once it lands.
Everything shifts earlier. A federal or statutory holiday midweek removes a processing day, so mail a day earlier than you normally would. The week between Christmas and New Year's is the extreme case: short-staffed facilities, weird pickup schedules, and backlogs. If a letter has to arrive during a holiday week, send it the week before and stop thinking about it.
